From appetite to flesh quality in rainbow trout: The role of ghrelin in regulating food intake and growth

The primary aim of the project is to clarify the regulatory role of ghrelin in food intake, body composition and growth performance in rainbow trout. Ghrelin is a newly discovered brain-gut peptide, and in mammals, data indicate that it represents a major regulatory link between food intake and growth regulation at the level of the CNS, as well as being a fattening hormone. The project mainly comprises studies of the role of ghrelin and its mechanisms of action in the following processes: (1) Appetite regulation and energy balance at different energy status; (2) Growth and deposition of fat in different tissues; (3) Secretion of GH and IGF-I, the primary growth-stimulatory endocrine system. Studies on the distribution, dynamics and characterization of the ghrelin receptor will also be done. The methodology involves central and peripheral homologous ghrelin administration, a validated ghrelin radioimmunoassay for rainbow trout, immunohistochemistry using validated ghrelin and ghrelin receptor antibodies, in situ hybridization, QPCR and enzyme kinetic analyses. The project will improve the understanding about basic endocrine processes regulating appetite, and growth, and how energy balance is achieved. In addition, the project has a great potential for an applied relevance for aquaculture fish production as it will generate integrated knowledge about growth and fattiness regulation, which can be used to optimize growth and flesh quality in farmed salmon.
