Taking sing carp as an example, it will be shown how the storage of frozen cell material can contribute to the preservation and survival of aquatic genetic resources and how a transfer of the concept to other species can be stimulated. Since the storage of somatic cells is not sufficient for the preservation of an animal population, and no techniques are yet known to allow the cryopreservation of fish egg cells in vital form, only the nuclei of fertilized and unfertilized ova and whole sperm should be stored. After thawing, the egg nuclei can then be replanted in enucleated ova from normal carp. Existing cryopreservation protocols on carp sperm are tested and possibly adapted. Fertilized and unfertilized oocytes from carp are gutted and these nuclei are cryopreserved. The seeds are then thawed again and placed in freshly pitted ova from another carp line. Larvae are extracted from the frozen material to confirm successful preservation. Then the appropriate breeders are approached in order to preserve sperm and egg nuclei from their endangered carp strains at the Fraunhofer EMB. In order to preserve a broad genetic diversity, as many strains as possible are to be used for cryopreservation. At least 25 individual samples per line are to be stored on sperm and nuclei.
