Methodology for genetic certification of aquaculture sturgeon species in order to increase the value of sturgeon products and for participation in restocking programs

The current projects aims at developing a validated methodology of genetic certification for aquaculture sturgeon stocks. The genetic test represents a frequent demand addressed by the potential buyers to the fish farmers and economic agents involved in processing the products coming from sturgeons. The necessity for molecular diagnosis of the species is motivated by the existence of the numerous attempts of frauds, mainly related with the caviar substitution. A correct identification of the species is complicated by the easiness with which the sturgeons hybridize giving birth to alive and sometimes fertile hybrids. This hampers the use of methods based on analysis the mitochondrial markers that are frequently assessed for species identification (e.g, DNA barcoding). The implementation of modern technologies such as Next Generation Sequencing allowed the sequencing of transcriptome in several sturgeon species and now the DNA sequences are available in public databases. This fact opens a new perspective for species identification based on nuclear markers sequencing and SNPs (Single Nucleotide Polymorphism) identification. These markers have a Mendelian codominant inheritance that allows the proper differentiation between the pure species and interspecies hybrids. Within the project the best panel of DNA markers (microsatellites and SNPs) allowing the correct molecular diagnosis and having a good cost per test will be designed and validated. Furthermore, a database comprising the results of the genetic assay will be organized and for the analyzed individuals a morphogenetic certificate will be elaborated. The previous collaboration between the partners and the significant scientific and technical expertise in the field clearly represent an advantage in ensuring the success of methodology elaboration, which may subsequently be used in order to help the marketing of the products by the economic operators at a correct price.
