Virus diseases constitute a major challenge for the Norwegian fish farming industry causing substantial economic losses every year. In order to reduce the negative effects of these diseases, more knowledge on the interaction between virus and host is necessary, to improve fish health through better management regimes and more efficient vaccines. Many research groups have demonstrated that miRNAs are pivotal in both adaptive and innate immunity in vertebrates. This project will provide new knowledge on miRNAs and their role in host pathogen interaction following virus infection in A. salmon. Host and virus-encoded miRNAs may be involved in resistance or viral encoded reprogramming of the host cell and may be important participants of the immune system. Analysis of differentially expressed miRNAs and target genes in challenge materials will point out genes that are directly involved in resistance/cellular reprogramming, and identify downstream miRNA genes that regulate immune system gene pathways. We have developed a pipeline analysis of Atlantic salmon miRNAs using modern genomic techniques. Our findings and pipeline will be utilized to carry out three research tasks: To characterize the miRNAs associated with -SAV infection and mortality. -IPNV infection and heritable difference in resistance. -To characterize the target genes modulated by the miRNA genes responding to viral infections (SAV, IPNV and ISAV). The project will contribute new knowledge about miRNA as participants in the immune system, their role in virus disease and disclose molecular details of disease mechanism that may aid in developing novel vaccines. The studies of our particular challenge materials may identify new and useful biomarkers (for marker assisted selection) and reveal miRNAs associated with and possibly causing, differences in mortality and disease resistance.
